effects of a voltage sensitive calcium channel blocker and a sodium-calcium exchanger inhibitor on apoptosis of motor neurons in adult spinal cord slices
Authors
abstract
objective: the apoptosis of motor neurons is a critical phenomenon in spinal cord injuries. adult spinal cord slices were used to investigate whether voltage sensitive calcium channels and na+/ca2+ exchangers play a role in the apoptosis of motor neurons. materials and methods: in this experimental research, the thoracic region of the adult mouse spinal cord was sliced using a tissue chopper and the slices were incubated in a culture medium in the presence or absence of n/l type voltage sensitive calcium channels blocker (loperamide, 100 μm) or na+/ca2+ exchangers inhibitor(bepridil, 20 μm) for 6 hours. 3-(4, 5-dimethylthiazol-2-yl)-2, 5 diphenyl tetrazolium (mtt) staining was used to assess slice viability while morphological features of apoptosis in motor neurons were studied using fluorescent staining. results: after 6 hours in culture, loperamideand bepridil not only increased slice viability, but also prevented motor neuron apoptosis and significantly increased the percentage of viable motor neurons in the ventral horns of the spinal cord. conclusion: the results of this study suggest that voltage sensitive calcium channels and na+/ca2+ exchanger might be involved in the apoptosis of motor neurons in adult spinal cord slices.

full textRole of a voltage-sensitive calcium channel blocker on inhibition of apoptosis in sensory neurons of cultured dorsal root ganglia in adult rat
Introduction: Under pathological conditions, abnormal increase in intracellular calcium concentrations is believed to induce cell death. In the present study, a voltage-sensitive calcium channel blocker (loperamide hydrochloride) was used to investigate its role in inhibition of apoptosis in sensory neurons of cultured spinal dorsal root ganglia (DRG). Methods: L5 DRG from adult rats were di...
full textEvaluation of Mutagenicity of Mebudipine, a New Calcium Channel Blocker
Mebudipine is a new dihydropyridine calcium channel blocker, synthesized in our laboratory, for treatment of hypertension. It has shown a better efficacy than other drugs in this group. For assessing the risks of this drug, certain safety tests in the preclinical stage have been performed. In this study mutagenic effect of mebudipine was evaluated using Ames assay that could assess the mutageni...

full textProtective effect of curcumin and Curcuma longa extract on apoptosis of motor neurons in cultured spinal cord slices of adult mouse
Introduction: Since Curcuma longa extract and curcumin have been shown to be potent antioxidant agents, they were used in cultured adult mouse spinal cord slices to investigate whether they can inhibit apoptosis in motor neurons. Methods: Slices from the thoracic region of adult mice spinal cord were divided into four groups: 1. Freshlyprepared slices (time 0) 2. Control 3. Slices treated by cu...
full textcaspase-dependent apoptosis in motor neurons of adult mouse spinal cord slices
in this study organotypic adult spinal cord slices were used to investigate whether caspases could participate in the apoptosis of motor neurons. the thoracic region of spinal cord was sliced using a tissue chopper and cultured in a medium for 6h. morphological and biochemical features of apoptosis were assessed by fluorescent staining and terminal deoxynucleotidyl nick end labeling (tunel) met...
